I'm Karloff would have been wonderful, but I suspect that Raines was the better choice for the role.

The Invisible Man is by far one of my favorite Universal films (it is also one of my favorite books), and even though Boris Karloff could have pulled it off, I don't think he could have delivered the way Rains did.
His dramatic body language accompanied by his voice is unmatched, and I wonder if this film would have been the same without Rains at the helm.
The FX still hold up well, and I can only imagine what the audience thought when it was released...they must have been astonished at this movie magic.

Amen, Brother! 
Rains' voice was, in a way, the star of the show. Even when he was not visible on screen, his perfect diction and his range of emotions created an acting tour de force.
And when he was all bandaged up and wearing the dark glasses, his vocal performance succeeded in doing what an actor's facial expressions usually do.
